About SLU-PP-332
SLU-PP-332 is an agonist of the estrogen-related receptors, mostly ERR-alpha, developed at Saint Louis University. ERRs control genes for mitochondria and fat burning in muscle.
It’s sometimes called an exercise mimetic.
ERR-alpha, ERR-beta and ERR-gamma are nuclear receptors that switch on genes for mitochondrial growth and fatty acid burning. Exercise turns that program on in muscle.
What is Orforglipron?
Orforglipron is a small molecule, not a peptide, that activates the GLP-1 receptor. It was discovered by Chugai Pharmaceutical and licensed to Eli Lilly, which developed it as a once-daily pill.
Because it isn’t a peptide, it doesn’t need the absorption enhancers that oral semaglutide relies on. It can be taken without food or water restrictions in trials.
Orforglipron binds a pocket in the GLP-1 receptor that differs from where the natural hormone binds, but it switches on the same signal. It leans toward G-protein signaling over beta-arrestin recruitment.
Oral research with SLU-PP-332
SLU-PP-332 is a small molecule. Most published mouse work injected it, so oral delivery is a newer research question.
Keep in mind that digestion breaks down many peptides, so a capsule study measures something different from a study using a reconstituted vial. Treat the two formats as separate variables.

What the research says
Billon and colleagues reported in 2023 that it raised endurance and fat oxidation in mice.
Later mouse work looked at obesity and heart function. Data are preclinical.
A phase 2 trial in adults with obesity, published in the New England Journal of Medicine in 2023, reported average weight loss of up to about 15% at 36 weeks.
Lilly reported phase 3 results from the ACHIEVE and ATTAIN programs in 2025, covering blood sugar control in type 2 diabetes and weight loss in obesity.
