What is FOXO4-DRI?
FOXO4-DRI is a modified peptide that blocks the interaction between FOXO4 and p53. It’s built from D-amino acids in reverse order, which makes it resistant to enzymes.
It’s studied as a senolytic, meaning a compound that clears senescent cells.
How FOXO4-DRI works
Senescent cells have stopped dividing but don’t die. In them, the FOXO4 protein holds p53 in the nucleus, which keeps p53 from triggering cell death.
FOXO4-DRI is a decoy that competes with FOXO4 for p53. With p53 released, it moves to the mitochondria and the senescent cell dies. Healthy cells barely use the FOXO4-p53 link, so they’re largely spared in the mouse studies.

Published FOXO4-DRI studies
Baar and colleagues reported in Cell (2017) that FOXO4-DRI pushed senescent cells into apoptosis and restored fur density, fitness and kidney function in aged and fast-aging mice.
A 2020 study tested it on senescent Leydig cells in aged mice.

How to reconstitute FOXO4-DRI 10mg
Here’s the math for 10mg of powder:
- 1mL of bacteriostatic water gives 10mg/mL
- 2mL of bacteriostatic water gives 5mg/mL
- 3mL of bacteriostatic water gives 3.33mg/mL
Go with whichever number is easiest to work with in your protocol, and label the vial with the concentration and the date you mixed it. The vial itself holds 3mL, so that’s the upper limit.
Bacteriostatic water is the usual choice. Add it slowly so it runs down the side of the vial, then swirl or roll until everything dissolves. Shaking can shear peptide chains. Skip it.
Storing FOXO4-DRI 10mg
Freeze-dried FOXO4-DRI 10mg handles room temperature fine during shipping, but for anything longer keep it in a freezer at -20°C, away from light. Once mixed, keep it in the fridge at 2 to 8°C and use it within about four weeks.
